Elicitation of immune responses against Nipah virus by an engineered synthetic DNA vaccine

چکیده
Nipah virus (NiV) is a re-emerging pathogen that causes severe disease in animals and humans. Current treatment measures for NiV infection are insufficient, there no approved vaccine against either humans or animals. listed as high-priority therapeutic research by the World Health Organization (WHO). In present study, we employed synthetic enhanced DNA technologies developed to design produce novel consensus Fusion (NiV-F) Glycoprotein (NiV-G) antigen sequences inclusion vaccines NiV. The expression of each was confirmed vitro using immune-binding assays. Electroporation-enhanced intramuscular injection NiV-F NiV-G into mice induced potent cellular immune responses multiple epitopes included antigen-specific CD8 + T cells. Both elicited high antibody titers mice, with single immunization sufficient seroconvert 100% immunized Additionally, also antibodies neutralize NiV-F-pseudotyped particles. These data support further study these nucleic acid-based antigens potential components an effective virus.

Enhanced Immune Responses of a Hepatitis C Virus core DNA Vaccine by co-Inoculating Interleukin-12 Expressing Vector in Mice
Background: Hepatitis C (HCV) is a worldwide problem without an effective vaccine with more than 170 million chronically infected people worldwide. DNA vaccines expressing antigenic portions of the virus with adjutants have recently been developed as a novel vaccination technology. Objectives: In the present study, a DNA vaccine expressing HCV core protein was developed with IL12 as a genetic a...
متن کاملRecombinant Measles Virus Vaccine Expressing the Nipah Virus Glycoprotein Protects against Lethal Nipah Virus Challenge
Nipah virus (NiV) is a member of the genus Henipavirus, which emerged in Malaysia in 1998. In pigs, infection resulted in a predominantly non-lethal respiratory disease; however, infection in humans resulted in over 100 deaths. Nipah virus has continued to re-emerge in Bangladesh and India, and person-to-person transmission appeared in the outbreak. Although a number of NiV vaccine studies have...
متن کاملAn Endogenous Immune Adjuvant Released by Necrotic Cells for Enhancement of DNA Vaccine Potency
Background: Improving vaccine potency in the induction of a strong cell-mediated cytotoxicity can enhance the efficacy of vaccines. Necrotic cells and the supernatant of necrotic tumor cells are attractive adjuvants, on account of their ability to recruit antigen-presenting cells to the site of antigen synthesis as well as its ability to stimulate the maturation of dendritic cells. Objective: T...
